Specimen: Blood

Container: White PPT - Plasma Preparation tube (preferred) or Lavender (EDTA). A separate tube is required if multiple tests are ordered and collected in a lavender tube. Microtainers are unacceptable.

Volume: 5 mL; minimum 3 mL

Unacceptable Conditions:

Heparinized specimens or specimens exposed to repeated freeze/thaw cycles.

Storage Instructions:

Store whole blood at 2-25°C for no longer than six (6) hours. For PPT, spin and refrigerate. For Lavender, spin, aliquot plasma and freeze. Ship specimen by overnight delivery, frozen on dry ice.

Turnaround Time (TAT):

Results reported in 7-11 days.

Test Availability:

Once per week; Results available on Fridays. Observed holidays may adjust the testing schedule.

Interpretive Data:

HIV-1 genotyping provides antiretroviral susceptibility information for protease inhibitors (PI) and reverse transcriptase inhibitors (NRTI, NNRTI) inhibitors. Genotyping should only be performed on blood samples with a known positive HIV-1 Subtype B viral load >500 copies/mL performed within 30 days of the requested genotype. This test is not intended to be used as a screening test or a confirmatory diagnostic test. From extracted RNA, the HIV-1 Genotyping System is used to generate a 1.8 kb reverse transcription-PCR amplicon.  The HIV-1 Genotyping System uses seven sequencing primers to create a consensus sequence of approximately 1.3 kb encompassing the entire Protease and 2/3 of the Reverse Transcription regions of the HIV-1 pol gene.
